A Method of Sterile Cotton Seedling Culture Characterised by Accelerating Germination with Wet Paper

Sterile Cotton Seedling Culture is the precondition of cotton tissue culture and Agrobacterium mediated gene transfer. It is difficult to get well seedling, for covering with so many busses and hard endothelium and having a great deal of bacterium and fungus. A new method of sterile cotton seedling culture is established in this study with Hengwu-8930 as testing material. High germination percentage with the filter paper, and taking easily off the seed coat, and low rate of abnormal plantlet are its characteristics. Brief course as next: the cotton seeds whose fuss are delinted with oil of vitriol were marinated in 0.1% HgCl2 for 20 minutes, then the surface-sterilized seeds were washed in sterile distilled water for 6 times. The seeds were germinated in dark between wet filter paper in sterilized petri plate at 25 °C for 2 days. After taking off the seed coat, the seedling were transplanted in 1/2 MS medium by the way of its radicale inserted in solid medium and grown in dark at 25 °C for 5 days.
